Predicted plasma protein binding (PPB) indicates how much drug is unbound (free) vs bound in circulation. Highly bound compounds may have altered pharmacokinetics. Percepta can provide PPB estimates as an early guide.
Solution
- Run the Protein Binding prediction module in Percepta for each compound.
- Interpret results qualitatively:
- Very high predicted binding may lead to low free concentrations but can also prolong half‑life.
- Very low binding increases free fraction but may affect exposure depending on clearance.
- Combine PPB predictions with other ADME properties (clearance, volume of distribution) for a holistic perspective.
- Use predictions to:
- Flag compounds for more detailed in vitro binding studies.
- Guide structural changes if extremely high or low binding is undesirable.
- Validate and refine interpretation rules as experimental PPB data become available.
