Description
After the user completes Screening and selects the best experiment, the selected condition and its processed data are passed to the Optimization wave (provided the strategy includes an optimization step). At this stage, the program is expected to recommend two additional gradient conditions, needed for reliable peak modeling. Instead of generated two recommendations, the user receives the warning: “Unable to calculate method parameters.”
Solution
This issue can be addressed in two ways.
Click Yes when prompted and manually create two additional gradient programs. Make sure these new gradients are different from the original condition, as AutoChrom needs variation between gradient programs to build a reliable peak model and perform optimization.
- Check that standard solvent and buffer names are used in the mobile phases.
AutoChrom may be unable to generate recommendations if it does not recognize the solvents or buffers defined in the project. This can happen when non‑standard or custom names are used (for example, entering “Meta” instead of a recognized name such as MeOH or Methanol).
Review the mobile phases in your AutoChrom project and ensure that only standard solvent names from the predefined list are used.
Note: For an existing project at this stage, solvent names cannot be edited, so this issue can only be avoided when setting up a new project.
