Description
Variable‑temperature (VT) NMR experiments show how spectra change with temperature, revealing dynamic processes (conformational exchange, hydrogen bonding). Spectrus can organize and compare such series.
Solution
- Acquire NMR spectra at the desired temperature points (e.g., from low to high temperature).
- Import all VT spectra into a single Spectrus project.
- Annotate each spectrum with its acquisition temperature in the metadata.
- Use stacked or overlay views to:
- Observe coalescence, line broadening, or splitting changes with temperature.
- Track chemical shift movements and peak intensities.
- For dynamic processes:
- Note temperatures where key changes occur (e.g., coalescence temperature).
- Use appropriate theoretical models (if needed) outside Spectrus to estimate rate constants.
- Include selected VT spectra and annotations in reports to document temperature‑dependent behavior.
