Spectrus: Using background subtraction to enhance low‑level components in LC/MS data

High chemical background or matrix signals can mask low‑abundance analytes. Spectrus allows background subtraction to enhance visibility of weak components.

Solution

1. Acquire an appropriate blank or matrix‑only run in addition to the sample runs.
2. Import both blank and sample LC/MS datasets into Spectrus.
3. For background subtraction:
   • Select the sample chromatogram or MS signal.
   • Use the Subtract function and choose the blank/matrix run as the background.
4. Adjust scaling or alignment so that general background is removed without over‑subtracting analyte peaks.
5. Inspect the result:
   • Confirm that low‑abundance analyte peaks are more visible after subtraction.
   • Ensure that genuine analyte signals are not lost.
