Description
When an AutoChrom project enters the Optimization wave, the initial processed dataset—based on the original gradient conditions—is carried over from the Screening wave or generated and processed directly in Optimization wave, depending on the strategy. AutoChrom uses this data to recommend two new gradient conditions. Because the recommendations are based on the processing results of the current data, different users working on the same project may obtain different results, which can affect reproducibility.
Solution
Recommended gradient conditions depend on how the first dataset is processed. The algorithm primarily uses the first and last eluted peaks to adjust the gradient percentage. These peaks can be any detected peaks—relevant or irrelevant, named or unnamed (including peaks labeled as unknown or “?”), or rejected (invisible).
To reliably reproduce the recommended conditions, remove all undesired peaks and ensure that the same first and last peaks are consistently selected.
